I cannot change the value in the "relative path and file name" field. I have tried many valid file names and every time it fails with "Could not save the configuration for Traffic Manager profile 'ilink'."
I have tried disabling the endpoint I have tried Disabling the Profile.
Is this possible?

It is strange. Would you like to create a new TM profile to set the "Relative path and file name". I suggest you could try to create a new one and set this value. And base on my experience, whatever the status of Azure TM profiles, we always could set them "Relative path and file name" . Please try to create a new one and test it.

It should be possible to change the monitoring path at any time. The monitoring path must follow these validation rules.
As Will suggested, please try your configuration using a new Traffic Manager profile. if that works, then please raise a ticket with Support to investigate why your original profile can't be updated. If a new profile doesn't work, it seems your settings may be failing the validation rules in some way.

I had to delete the EndPoint and recreate it in the end.
There was nothing wrong with the configuration - it just wouldn't allow a change.
